自发性先天性白内障大鼠皮肤移植及生化位点的研究

摘要:目的 探讨近交系同系异体自发性先天性白内障( TSC)大鼠皮肤移植反应的影响以及生化标记基因位点的测定方法 选用 TSC 大鼠 10 ,6 ~ 8 周龄,体质量 170 ~ 200 g,雌雄各半,采用背-背皮肤移植法进行自体和异体皮肤移植,一周后拆除包扎,观察皮片生长情况取基础群 F29 代的同一窝群 TSC 大鼠 6 ,6 ~ 8 周龄,体质量200 ~ 220 g,雌雄各半按照中华人民共和国国家标准 GB / 14923—2010 实验动物哺乳类实验动物的遗传质量控制、GB / T 14927. 1—2008 实验动物近交系小大鼠生化标记检测法测定结果 皮肤移植 TSC 大鼠连续观察 100 d皮片成活生长。 11 个生化标记基因位点基因型为:酯酶-1 a,酯酶-3 a,酯酶-4 b,酯酶-6 b,酯酶-8 b,酯酶-9 c,酯酶-10 a,血红蛋白 β 链为 b,过氧化氢酶-1 a,碱性磷酸酶-1 a,血清碱性磷酸酶-1 b,参照目前常用的近交系大鼠品系的生化遗传概貌,未发现与 TSC 大鼠的生化标记基因位点完全相同的,TSC 大鼠具有独特的遗传特征结论 皮肤移植无排斥,说明 TSC 大鼠的组织相容性基因是一致的;酯酶-1 11 个生化标记位点结果均为纯合型,符合近交系质量标准规定

Abstract:

Abstract: Objective To investigate the anti-rejection effect of xenogenic skin transplantation and biochemical marker genes in congenital cataract rats ( TSC) . Method Ten male TSC rats (170-200 g) were chosen for tergal skin transplantion of the autologous and allogeneic. One week later, bandage was removed and skin groeth was observed. We chose 6 TSC rats (200-220 g) from the same nest of basic group F29 generation, half male and half female, aged from 6 - 8 weeks. Tests were measured according to Chinese national standards GB / 14923—2010( laboratory animal-genetic quality control of mammalian laboratory animals) and GB / T 14927. 1—2008 ( laboratory
animals-method for biochemical markers of inbred mice and rats) . Result The skin graft survival with conclusious observation of 100 days from TSC rats. The marker loci were Akp1 a, Cs1 a, Es1a, Es3 a, Es4 b, Es6 b, Es8 b, Es9 c, Es10 a, Hbb b and Alp1 b, respectively. Referring to the biochemical genetic profile of the inbred rat strains commonly used at present, we did not find exactly the same biochemical markers with our TSC rats. Our TSC rats have unique genetic profiles. Conclusion There was no rejection after skin transplantation, which indicated that the histocompatibility gene of inbred TSC rats was consistent. The result of 11 biochemical markers
such as esterase-1 were homozygous, which met the quality standards of inbred lines.
