摘要: 摘要: 目的评价改良的大鼠经皮电阻试验方法的可行性。方法取28 ~ 30 日龄大鼠背部皮肤制成直径约20 mm的皮片,筛选电阻值大于20 kΩ 的皮片用于试验。8 个受试物,每个受试物设3 个平行样,分别取150 μL 样品加至皮片表皮层,作用24 h。然后洗去受试物,测定皮片电阻值,对电阻值≤15 kΩ 的皮片进行罗丹明B 染色、提取、565 nm测定吸光度值,计算皮片染料吸收量,进而判定受试物的腐蚀性。结果与传统动物试验方法结果比较,改良的大鼠经皮电阻试验方法灵敏度为100%、特异度83%、准确度88%,均符合经济合作与发展组织对该方法的要求。结论改良的大鼠经皮电阻试验方法在本实验室是可行的。

Abstract: Abstract: Objective To assess modified in vitro transcutaneous electrical resistantce test method. Method 28 ~30 day-old rats are humanely killed,the dorso-lateral skin of each animal is then removed. Skin discs with a diameter of approximately 20 mm each,are removed. If the resistance value is greater than 20 kΩ,the discs could be used for the test method. Test chemicals ( 150 μL) are applied uniformly to the epidermal surface. Three skin discs are used for each test. Test chemicals are applied for 24 hours at room temperature,and then are removed by washing. The skin impedance is measured as TER. If TER values produced by such chemicals are less than or around 15 kΩ,an assessment of dye penetration should be carried out. The optical density ( OD) of the incubation solution is measured at 565nm. The sulforhodamine B dye content per disc is calculated from the OD values. Result The sensitivity,specificity and accuracy of the modified TER test method are 100%,83% and 88% respectively. The TER test method is considered valid to discriminate corrosive from non-corrosive chemicals.Conclusion The modified TER test method is valid in our Lab.
