The Decontamination of Rag2 Knockout Rats by Caesarean Section

摘要: 摘要: 目的 利用剖腹产的方法对基因工程 Rag2 敲除大鼠进行净化。方法 分别对见栓第 20 天和第 22 天的孕鼠 进行剖腹产手术,剖得仔鼠于 6—8 日龄时进行剪尾作 PCR 基因型鉴定,6—8 周龄进行微生物检测和基因型鉴定。 结果 大鼠剖腹产于见栓第 22 天进行,可以显著提高成活率,平均离乳率 84. 48% ; 净化后经检测,微生物等级符 合 SPF 级标准; 经基因型鉴定,可获得纯合子大鼠。结论 Rag2 敲除大鼠通过剖腹产净化可获得 SPF 级纯合子后 代,证明该方法可行。

Abstract: Abstract: Objective To decontaminate Rag2 knockout rats by caesarean section. Method The caesarean section was taken on the 20th and 22th day after vaginal plugs detection,the pups were genotyped by PCR after 6—8 days of birth,and pathogens were tested at 6—8 weeks. Result The survival rate was significantly increased when the operation took on the 22th days after vaginal plugs detection,the average rate of weaning is 84. 48%. Pathogens test results met the National SPF standards of experimental animal. Genotyping results were positive,and got homozygote offspring after one time of cross. Conclusion SPF homozygote offspring of Rag2 knockout rats were achieved by caesarean section successfully.
