使用eCCI 仪制备大鼠TBI 动物模型及血清S100B 和NSE 含量变化

The Study on TBI Model Praperation with eCCI in Rats and the Change of Serum S100B and NSE Content

摘要: 摘要: 目的为科学研究提供重复性好、损伤( TBI) 程度稳定的颅脑损伤( TBI) 动物模型制备方法。方法选用SD 大鼠,随机分为正常A 组、模型B 组、模型C 组、模型D 组和假手术E 组。使用电子颅脑损伤仪( eCCI) ,选定不同 打击参数分别对模型B 组、模型C 组、模型D 组打击造模,制备不同损伤程度的TBI 模型,通过观察血清S100B 和 NSE 含量变化评价模型制备效果。结果损伤发生后第1 天、第2 天、第3 天血清S100B 和NSE 含量均呈上升趋 势,模型组与假手术组比较均有显著性差异( P < 0. 01) ; 各模型组之间表现为随打击力度的增加,S100B 和NSE 含 量显著性增高( P < 0. 01) 。结论利用eCCI 仪制备TBI 大鼠模型,能够通过打击速度、打击深度参数的设置而精 确控制模型的损伤程度,具有易操作、打击强度可控制、重复性好等优点,能够根据研究需要制备各种理想损伤程 度的动物模型

Abstract: Abstract: Objective To study the TBI rats model produced by eCCI and provide a model establishing method of easy operation,the controllable degree of damage,good repeatability for the TBI experimental study. Method 65 SD rats,which were randomly divided into blank control group A,model group B,model group C,model group D and sham-operation group E,were produced as the animal model of brain injury in different degree by eCCI according to different selected parameters. The speed was 3 m/s and the depth was 3 mm in model group B. The speed was 4 m/s and the depth was 3 mm in model group C. The speed was 5 m/s and the depth was 3 mm in model group D. The quality of the model was evaluated through detecting the dynamic changes of S100B and NSE by ELIASA in animal serum. Result The content of S100B and NSE in serum was increased on the first day, second day,third day after animal damaged in model group by ELISA. Between the model group and sham operation group,there were significantly different ( P < 0. 01) . In model group B,C and D,the content of S100B and NSE was increased significantly with the increase of hit ( P < 0. 01) . Conclusion By using eCCI TBI rat model can be established with regulation of hit speed and depth. The damage of model was related to the hit speed. The method of TBI model preparation has the advantages of easy operation and control,better repeatability,and can be used according to the need to make various animal model for the study.
