Optimization of the Proliferation of Mouse Spleen T Lymphocytes

摘要: 目的 探讨刀豆球蛋白 A 刺激小鼠脾 T 淋巴细胞增殖的最佳浓度与最佳作用时间。 方法 以小鼠脾 T 淋巴细胞与刀豆球蛋白 A 共同培养后,采用 CCK-8 法检测淋巴细胞的增殖情况。 结果 不同浓度的刀豆球蛋白 A 对小鼠脾 T 淋巴细胞的刺激效果不同,在作用后 48 和 72 h 检测,10 ~ 25 μg / mL 浓度范围内增殖最为明显。 结论 刀豆球蛋白 A 对小鼠脾 T 淋巴细胞的增殖具有双重作用,推荐浓度为 10 ~ 25 μg / mL,检测时间为 48 ~ 72 h。

Abstract: Objective This study was aimed to explore the appropriate concentration and stimulation duration of concanavalin A in the T lymphocyte proliferation test of mouse spleen. Method The mouse spleen T lymphocytes were cultured in vitro without or with concanavalin A at different concentrations, the proliferation of the T lymphocytes were analyzed with CCK-8 kit at different timepoint after culture. Result
The result showed different concentrations of concanavalin A had different effects on mouse spleen T lymphocyte proliferation, and the best proliferation effect was observed in the concentration ranged from 10-25 μg / mL with a 48 or 72 h stimulation duration. Conclusion In the mouse spleen T lymphocytes proliferation test, the recommended concentration for ConA is 10 - 25 μg / mL, and the recommended timepoint for analysis is at 48 to 72 h after invitro culture.
