熊果酸对H22 荷瘤小鼠抗肿瘤及免疫调节作用

Antitumor Effect and Immune Regulation Activity of Ursolic Acid on Hepatoma H22 in Mice

摘要: 摘要: 目的探讨熊果酸( UA) 对H22 荷瘤小鼠抗肿瘤作用及免疫功能的影响。方法皮下移植建立H22 荷瘤小 鼠模型,腹腔注射不同剂量UA,检测抑瘤率和免疫器官指数,MTT 法检测脾脏T、B 淋巴细胞增殖能力,流式细胞术 检测CD4 +、CD8 + T 细胞亚群含量及比例,ELISA 法检测血清细胞因子IL-2、IL-4 和TNF-α 的表达量。结果UA 对 小鼠皮下移植性肿瘤H22 有显著的抑制作用,可降低免疫器官中异常增大的脾指数,增强脾脏中T、B 淋巴细胞增 殖能力,提高淋巴细胞亚群CD4 + T 细胞表达及CD4 + /CD8 + T 细胞亚群比例,促进血清IL-2、TNF-α 表达,降低IL-4 表达。结论UA 可抑制小鼠肝癌H22 肿瘤生长,体内可以提高荷瘤小鼠的免疫能力,其抗肿瘤作用可能与机体的 免疫调节作用相关。

Abstract: Abstract: Objective The aim of this study was to investigate the antitumor effect and immune regulation activity of Ursolic acid ( UA) on H22 in mice. Method The models of tumor-bearing mice were established by i. p. injection of hepatic cancer H22 and the mice were randomly divided into 5 groups,each groups given by i. p. The tumor inhibitory rate,the thymus index and spleen index were calculated,lymphocyte proliferation of spleenstimulated by ConA and LPS were detected by MTT,T cell subsets CD4 + ,CD8 + were detected by FCM and the levels of The cytokine IL-2,TNF-α, IL-4 in serum were detected by ELISA. Result In vivo,the inhibitory rate of UA significantly higher than that of the model group. Compared with the model group,spleen index of high dose UA group were significantly lower. The T and B lymphocyte proliferation of spleen were significantly higher. The CD4 + T cell subset and CD4 + /CD8 + T rate were significantly increased,but CD8 + T cells has no significant change. The cytokine IL-2,TNF-α in serum expression were promoted with high dose UA,meanwhile the IL-4 expression was reduced. Conclusion The results show that UA can inhibit tumor growth both in vitro and in vivo. It could play a role of antitumor activity in vivo by regulating the body’s abnormal immune function in tumor-bearing mice.
