阿尔茨海默病发病机制及体内外模型研究进展

Research Progress in Pathogenesis and in Vitro-in Vivo Model of Alzheimer’s Disease

摘要: 阿尔茨海默病(AD)是一种以认知功能障碍为主要特征的神经退行性疾病,具体发病机制目前尚不清晰。 随着我国人口老龄化越来越严重,患有 AD 人群的比例越来越高,亟需深入探究其发病机制并建立合适的疾病模型。本文从当前 AD 的发病假说和体内外模型进行了综述,详细介绍了神经细胞 β-淀粉样前体蛋白( APP ) 转基因模型、D-半乳糖(D-gal)模型、D-半乳糖联合 β-淀粉样蛋白(Aβ)模型等 10 种体内模型和 3 种体外模型( HT22 细胞模型、PC12 细胞模型和 SH-SY5Y 细胞模型) ,以期为 AD 疾病的基础研究和新药开发提供模型参考和研究思路。关键词: 阿尔茨海默症;发病机制;体内动物模型;体外细胞模型

Abstract: Alzheimer’ s disease ( AD ) is a neurodegenerative disease characterized by cognitivedysfunction. As China’ s population aging becomes more and more serious, the proportion of peoplesuffering from AD is increasing, it is urgent to study its pathogenesis and establish appropriatepathological models to further study the prevention and treatment of AD. In this paper, in vitro and invivo AD models were reviewed, especially ten in vivo animal models (β-amyloid precursor protein (APP)transgenic model, D-galactose model, D-galactose combined with β-amyloid protein (Aβ) model and soon) and three in vitro models ( HT22 cell model, PC12 cell model and SH-SY5Y cell model) wereintroduced in detail. This study will provide AD reference and research ideas for further study and newdrug development for AD disease.
