和血明目片对激光诱导小鼠脉络膜新生血管的治疗作用

Therapeutic Effect of Traditional Chinese Medicine on a Mouse Model of Choroidal Neovascularization

摘要:

摘要:目的 观察和血明目片对激光诱导小鼠脉络膜新生血管( choroidal neovascularization,CNV) 是否具有治疗作用。 方法 选取 8 周龄雄性 C57BL / 6 小鼠为实验动物。 CNV 小鼠进行 577 nm Nd:YAG 激光诱导( 能量 180 mW、光斑 100 μm、曝光时间 100 ms、光斑数 6 个) ,产生脉络膜新生血管后随机分为生理盐水组、小剂量组、中剂量组和大剂量组,每日进行生理盐水与和血明目片灌胃。 对照组小鼠除激光诱导外,其余操作同实验组小鼠。 对照组与生理 盐 水 组 生 理 盐 水 灌 胃 10 mL / ( kg· d ) , 小 剂 量 组、 中 剂 量 组 和 大 剂 量 组 分 别 给 予 0. 34 g / ( kg· d ) 、0. 68 g / ( kg· d) 、1. 36 g / ( kg· d)和血明目片灌胃,在干预后 1 周进行视网膜电流图、眼底荧光造影检查和脉络膜铺片新生血管染色。 结果 视网膜电流图结果显示各剂量和血明目片干预组的暗视 3. 0 反应 b 波幅值均高于生理盐水组( P<0. 05) ,且与和血明目片剂量呈正相关。 活体动物眼底荧光造影结果显示中剂量组和大剂量组眼底血管渗漏程度明显轻于生理盐水组( P<0. 05) 。 离体脉络膜铺片测量新生血管面积结果与眼底荧光造影结果基本一致。 结论 本研究从功能和形态两个方面证实了和血明目片对 CNV 小鼠病理损伤 CNV 具有一定的治疗作用,为下一步开展相关机制研究奠定了基础。

Abstract:

Abstract: Objective To observe the therapeutic effect of hexuemingmu ( HXMM ) tablet on laser-induced choroidal neovascularization ( CNV) in mice. Method C57BL / 6 mice ( aged 8 weeks, male) were selected as experimental animals. CNV mice were induced by double frequence Nd: YAG laser ( wave length: 577 nm; power: 180 mW; duration: 100 ms; spot size: 100 μm; number of light spots: 6. After generation of choroidal neovascularization, CNV mice were randomly   divided into saline group, small dose group, moderate dose group and large dose group. The other operations of the control group were the same as those of the experimental groups except laser exposure. The control group and saline group were gavaged with 10 mL / kg per day of saline. The small dose group, moderate dose group and high dose group were given 0. 34 g / kg, 0. 68 g / kg, 1. 36 g / kg HXMM ( the drug was dissolved in saline) per day respectively. Electroretinogram, fundus fluorescein angiography and choroidal neovascularization staining were performed 1 week after intervention. Result Electroretinogram showed that the amplitude of scotopic 3. 0 response b wave of intervention groups with HXMM was higher than that of saline group ( P<0. 05) . The result of fluorescein angiography in vivo showed that the degree of fundus fluorescence leakage in moderate dose group and high dose group was significantly lower than that in saline group ( P<0. 05) . The result of area of neovascularization measurement was consistent with fundus fluorescein angiography. Conclusion This study confirmed that HXMM has a certain therapeutic effect on CNV mice from aspects of function and morphology, which laid a foundation for further research on related mechanisms.
