培达对STZ糖尿病大鼠血脂及血流变学指标影响的观察

Effects of Cilostazol on Blood Lipoids and Haemorheological Indexes in Rats With Diabetes Induced by Streptozotocin

摘要: 目的观察培达对实验性糖尿病大鼠血脂及血流变学指标的作用效果。方法8周龄的SD雄性大鼠24只随机分为3组:正常对照组(NC,n=8),糖尿病对照组(DC,n=8),和糖尿病培达干预组(DP,n=8)。实验性糖尿病经大鼠尾静脉注射链脲霉素STZ(45mg/kg)诱导。糖尿病培达干预组每日经灌胃给培达(50mg/kg)一次。治疗24周后取血测血甘油三酯和总胆固醇及血流变学指标。结果与正常对照组相比,糖尿病组大鼠血甘油三酯显著升高86.4%(P<0.05)。培达治疗可显著降低甘油三酯水平(P<0.05)。糖尿病对照组大鼠的水洗红细胞低切粘度(反映红细胞变形能力)显著增加(P<0.05)。培达治疗显著改善红细胞变形能力的作用。结论本实验显示,培达改善糖尿病大鼠红细胞的变形能力,降低血甘油三酯水平,加上抑制血小板聚集的作用,有可能成为临床预防糖尿病慢性并发症发生的研究药物。

Abstract: Objective\ To investigate the effects of cilostazol on blood lipoids haemorheological indexes in rats with diabetes induced by streptozotocin.Methods\ 24 eight_week_old male SD rats were randomly divided into normal control group,diabetes model group,and diabetes group treated by cilostazol,and there were 8 rats in each group.Experimental diabetes was induced by a sigle dose(45mg/kg) of injection of streptozotocin via the tail vein.After the onset of diabetes,cilostazol was orally administered into each rat in the treated group at 50mg/kg/day.Haemorheological indexes,plasm toatal cholesterol and triglyceride were determined after being treated by cilostazol for 24 weeks.Results\ As compared with normal control group,plasm triglyceride in diabetes model group was increased by 85%,and the treatment by cilostazol could produce a significant increase in the washed erythrocyte viscosity at low shear(an index of erythrocyte deformability) and a significant decrease in plasm triglyceride.Conclusion\ It was suggested that cilostazol could produce a significant increase in the washed erythrocyte viscosity at low shear and a significant decrease in plasm triglyceride in the diabetic rats.In addition,the inhibitory effect of cilostazol on platelet aggregation may provide a new perspective in the field of prophylaxis for chronic complications of diabetes.
