Effect of Genetic Contamination of BALB/c Mouse on the Preparation of Monoclonal Antibodies

摘要: 目的检测BALB/c小鼠遗传污染对单克隆抗体制备实验的影响。加强近交系小鼠的饲养管理和重视实验动物的遗传质量检测。方法使用遗传生化标记方法对购进的两批BALB/c小鼠进行了碱性磷酸酶-1(Akp-1)等13个生化位点的检测。结果注射杂交瘤细胞后不产生腹水,经生化检测发现,购买的第一批BALB/c小鼠,在过氧化氢酶-2(Ce-2)等4个生化位点发现杂合基因,第二批BALB/c小鼠在过氧化氢酶-2(Ce-2)等8个生化位点发现杂合基因。结论本实验说明在实验动物的饲养管理上存在漏洞,上述两批BALB/c小鼠未严格按照国家实验动物管理操作规程操作,必须规范实验动物饲养管理,加强实验动物遗传检测,这是保证动物试验结果准确可靠不可或缺的重要环节。

Abstract: Objective\ To monitor the influence of the genetic contamination of BALB/c Mouse on the preparation of monoclonal antibodies.To strengthen the management and breeding of the inbred strains mouse and improve the detection on the genetic quality.Methods\ The method of biochemical label was used to detect the 13 alleles(Akp-1,etc)of two groups of BALB/c mouse.Results \ After the injection of hybridomas cells,no ascites was produced in these of the BALB/c mouse.Four heterozygosis at the points of Ce-2 etc were found in the first group of mouse,and 8 heterozygosis at the points of Ce-2 etc were found in the second group of mouse.Conclusion\ The results proved that there were shortomings in management and breeding of the laboratory animals,and it is important to enhance the management about laboratory animals and strengthen the detection of genetic quality.
