Comparative Evaluation of Different Anesthesia Protocols in Cynomolgus Monkeys

摘要: 目的 比较不同麻醉方案在食蟹猴实验中的效果及安全性,为动物实验中麻醉管理提供优化策略。方法 将72只猴子(年龄为1~6岁和12~17岁)分为3组麻醉方案:(1)静脉注射3%戊巴比妥钠;(2)肌肉注射盐酸氯 胺酮(10 mg/kg);(3)氯胺酮诱导联合丙泊酚输注。系统监测了麻醉诱导时间、维持时间、核心生理参数(体温、 RR、HR)以及血清生化标志物(AST、ALT、CK)。结果 戊巴比妥钠在青少年猴子(1~6岁)中表现效果较好,诱导 迅速(≤2 min),维持时间较长(≥100 min),而中老年动物(12~17岁)需要谨慎调整剂量,表现出诱导时间延长且 维持时间缩短;单用氯胺酮提供了可靠的短期麻醉(10~20 min),对生理干扰最小。氯胺酮-丙泊酚组合对于长时 间麻醉有效,但需注意体温下降风险,尤其是术后生化变化(特别是CK升高)。结论 这些发现表明食蟹猴的麻 醉反应具有年龄依赖性,为非人灵长类动物实验的标准化麻醉管理提供了实验依据。

Abstract: Objective To systematically compare the efficacy and safety of different anesthetic protocols in Cynomolgus monkeys for optimizing anesthesia management in primate research.Methods Seventy two monkeys were stratified by age into juvenile (1-6 years) and senior (12-17 years) groups, receiving one of three anesthetic regimens: (1) intravenous 3% pentobarbital sodium; (2) intramuscular ketamine hydrochloride (10 mg/kg); or (3) ketamine induction followed by propofol infusion. Comprehensive monitoring included anesthesia induction time, maintenance duration, core physiological parameters (core temperature, heart rate, respiratory rate), and serum biochemical markers (AST, ALT, CK). Results Pentobarbital sodium demonstrated optimal performance in juvenile subjects, characterized by rapid induction period (≤2 min) and prolonged duration (≥100 min), whereas senior animals exhibited delayed induction period and reduced efficacy, necessitating dose adjustment. Ketamine monotherapy provided stable short-duration anesthesia (10-20 min) with minimal physiological perturbations. The ketamine-propofol combination proved effective for extended procedures but required careful monitoring of thermoregulation and postoperative biochemical alterations, particularly significant CK elevation.Conclusion This study reveals age-dependent anesthetic responses in M. fascicularis, establishing evidence-based guidelines for protocol selection in nonhuman primate research to enhance experimental standardization and animal welfare.
