Phenotypical Difference and Genetic Quality Control in C57BL/6 Substrains

摘要: C57BL/6小鼠是迄今为止应用最广泛的实验室啮齿类动物,其中C57BL/6J和C57BL/6N为两大核心谱系, 经长期繁育又各自衍生出多个亚系,亚系间的遗传背景差异易引发表型分化,进而直接影响实验结果的可靠性。 若选用遗传背景不明确的C57BL/6亚系开展实验,其潜在的遗传差异可能对研究结果产生不可预估的干扰。因 此,C57BL/6种群的繁育机构需重视并落实其遗传质控工作,通过科学措施及时发现遗传污染或亚系混杂问题,维 持亚系的遗传背景稳定与清晰,从源头杜绝遗传背景混杂对研究的干扰。

Abstract: The C57BL/6 mouse is the most widely used experimental rodent to date. C57BL/6J and C57BL/6N represent the major lineages of C57BL/6 mouse, from which a large number of substrains have been derived following long-term independent breeding. Genetic background differences among substrains are likely to cause phenotypic divergence, which directly affects the reliability and reproducibility of experimental result. If C57BL/6 substrains with unclear genetic backgrounds are used in experiments, potential genetic differences may cause unpredictable interference to the research result. Therefore, institutions responsible for breeding C57BL/6 mouse colonies should prioritize and implement genetic quality control. Scientific measures should be taken to promptly identify genetic contamination or substrain admixture, maintain the stable and well-defined genetic background of the substrains, and eliminate interference with research caused by mixed genetic backgrounds at the source.
