 A Method for Establishing a Rat Model of Osteoporosis Combined with Hyperlipidemia

摘要: 目的 本研究建立一种骨质疏松合并高脂血症大鼠模型,为今后研究骨质疏松症合并高脂血症的发病机制 和药物有效性提供动物疾病模型选择的依据。方法 12只雄性SPF级大鼠适应性喂养一周后,随机分为对照组6 只和实验组6只。对照组给予普通饲料,实验组给予60%高脂饲料加肌肉注射地塞米松(5 mg/kg,每周 2 次),造 模13周后取材。计算肝系数、体脂率;全自动生化仪检测血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白 (LDL)、高密度脂蛋白(HDL);取部分肝组织进行HE染色并观察其病理形态,剖取右侧股骨,进行骨密度检查。 结果 与对照组相比,实验组大鼠肝系数显著升高(P<0.01),体脂率显著升高(P<0.01);TG、TC、HDL、LDL含量 均显著升高(P<0.05);肝表面颜色变黄,边缘变钝,质地较软;大鼠肝细胞排列紊乱,体积明显增大,出现大面积脂 肪变性。细胞质内有较大脂滴空泡,部分细胞核受其挤压,被推至细胞边缘;右股骨R1区、R2区和右股骨全部区 域骨密度含量均显著下降(P<0.001)。结论 通过选用高脂饲料喂养联合肌肉注射地塞米松给药13周能成功建 立骨质疏松合并高脂血症大鼠模型。

Abstract: Objective This project aims to establish a rat model of osteoporosis combined with hyperlipidemia, providing a basis for selecting animal disease models for future studies on the pathogenesis and therapeutic efficacy of osteoporosis complicated with hyperlipidemia.Methods After one week of adaptive feeding, 12 rats were randomly divided into a control group with 6 rats and an experimental group with 6 rats. The control group was given regular feed, while the experimental group was given 60% high-fat feed plus intramuscular injection of dexamethasone (5 mg/kg, twice a week), after 13 weeks of modeling, samples were taken. Calculate liver coefficient and body fat percentage.The fully automated biochemical analyzer detects serum levels of total cholesterol (TC), triglycerides (TG), low-density lipoprotein (LDL), and high-density lipoprotein (HDL); part of the liver tissue was harvested for HE staining and observation of its pathological morphology, and the right femurs were dissected for bone density examination.Results Compared with the control group, the liver coefficient and body fat percentage of rats in the experimental group were significantly increased (P<0.01 and P< 0.01, respectively).The levels of TG, TC, HDL, and LDL were significantly increased (P<0.05, respectively). The surface color of the liver turns yellow, the edges become dull, and the texture is softer; the arrangement of rat liver cells is disordered, with a significant increase in volume and extensive steatosis. There are large lipid droplets and vacuoles in the cytoplasm, and some nuclei are compressed and pushed to the edge of the cell.The bone density content in the right R1, right R2, and all right regions significantly decreased (P<0.001, respectively).Conclusion A rat model of osteoporosis combined with hyperlipidemia can be successfully established by feeding with high-fat diet and administering dexamethasone via intramuscular injection for 13 weeks.
