不同配方高脂饲料诱导建立大鼠高脂血症模型的比较研究

Comparative study on the establishment of hyperlipidemia models in rats induced by different high-fat diets

摘要: 目的 应用不同配方的高脂饲料,诱导建立 Sprague-Dawley( SD)大鼠高脂血症模型。 方法 雄性 SD 大鼠随 机分成 6 组:正常对照组和 5 个高脂模型组,连续分别饲喂普通饲料和不同配方高脂饲料 6 周,于诱导 1、2、4 和 6 周检测大鼠血脂含量,诱导 6 周后进行肝脂质含量的检测。 结果 与正常对照组大鼠相比,造模 6 周后,模型 2 和 模型 3 组大鼠血清中总胆固醇( TC)含量显著升高,各模型组大鼠血清中的低密度脂蛋白-胆固醇( LDL-C) 含量均 显著升高;且与正常组大鼠相比,各模型组大鼠肝中的甘油三酯( TG)和总胆固醇( TC)含量也显著增加。 结论 通 过饲喂高脂饲料 6 周进行诱导,添加了 20%果糖和 10%猪油的配方 2 和配方 3 高脂饲料能够成功建立大鼠高胆固 醇血症模型,为降脂药物的研发提供了一种简单、低成本、稳定的动物模型。

Abstract: Objective To discover the hypolipidemic drugs, we established the model of hyperlipidemia in Sprague-Dawley ( SD) rats by feeding the high-fat diets of different recipes. Method Male SD rats were randomly assigned to 6 groups:the control group and 5 model groups. The rats were fed with the control diet and the high-fat diet of five different recipes for 6 weeks respectively. The concentrations of serum lipids were detected at the end of 1, 2, 4 and 6 weeks ’ feeding. The hepatic lipids ’ concentrations were determined at the end of 6 weeks. Result Compared with the control group, after 6 weeks of feeding, the concentrations of serum total cholesterol ( TC) of the rats in model group 2 and 3 significantly increased; the concentrations of serum low-density lipoprotein cholesterol ( LDL-C) of the rats in all model groups significantly increased. And the hepatic lipids ’ concentrations significantly increased in all model groups. Conclusion The model of hypercholesteremia was established by feeding the high-fat diet of recipe 2 and 3 that adding 20% fructose and 10% lard for 6 weeks in rats. this experiment provides a simple, low-cost, and stable animal model for the development of lipid-lowering drugs.
